mutations of basal core promoter and precore regions in hepatitis b virus genotypes b and c
نویسندگان
چکیده
conclusions the mutation ratio difference between genotypes b and c in children was higher than that of adults and several combined mutations were exclusively detected in children with chronic hbv genotype c infection associated with higher viral load. objectives the aim of this study was to assess the mutation profiles of bcp and precore regions in different hbv genotypes in chronically infected children. patients and methods a cohort of 245 children and 92 adults with chronic hbv infection was included in this study. bcp and precore regions were analyzed by pcr amplification and sequenced. results ten nucleotide positions, including nt1679, nt1721, nt1753, nt1757, nt1758, nt1762, nt1764, nt1775, nt1856 and nt1858 in bcp/precore regions of hbv genome, showed obviously higher frequencies of mutation in genotype c subjects than in genotype b subjects among children, while there were only three positions, including nt1679, nt1758 and nt1775 showing higher mutation frequencies in genotype c subjects than in genotype b subjects in adults. several combined mutations were obviously highly distributed in children with chronic hbv genotype c infection, such as g1721a/a1775g/t1858c triple mutation; a novel combined mutation type, exclusively detected in children with chronic hbv genotype c infection. in addition, g1721a/a1775g/t1858c combined mutation was associated with higher viral load and lower age distribution. background mutations in basal core promoter (bcp) and precore regions of hepatitis b virus (hbv) are associated with course and treatment outcomes of chronic hbv infection. while bcp and precore mutation analysis have been carried out in adult patients between different genotypes, this analysis has rarely been performed for chronically infected children.

Mutations of Basal Core Promoter and Precore Regions in Hepatitis B Virus Genotypes B and C
OBJECTIVES The aim of this study was t...
متن کاملMutations in pre-core and basal-core promoter regions of hepatitis B virus in chronic HBV patients from Golestan, Iran
Objective(s): It has been reported that the mutation of the pre-core (PC) and basal-core promoter (BCP) may play an important role in the development of HBV-related hepatocellular carcinoma (HCC). In this study the PC and BCP mutations were investigated in chronic HBV patients. Materials and Methods:In this study, 120 chronic HBV patients from Golestan, Northeast of Iran who were not vaccinated...
متن کاملMutations in the Basal Core Promoter and Precore/Core Regions of Hepatitis B Virus in Patients Co-Infected With Human Immunodeficiency Virus
ABSTRACT Background and objectives: Globally, about one third of the population has been infected with Hepatitis B virus (HBV) and more than 400 million people have become chronically infected. Nearly, 20-25% of all carriers develop serious liver diseases such as cirrhosis, chronic hepatitis and hepatocellular carcinoma (HCC). According to t...
متن کاملHepatitis B virus core promoter and precore mutations and their relatedness to genotypes and disease pathogenesis
Despite the successful vaccination programme worldwide, Hepatitis B virus infection remains a global problem. Infection rate in developing countries is higher in comparison to the developed countries. Core promoter region (nts.1591-1822) plays a very important role in HBV replication, morphogenesis and pathogenesis, whereas the precore region (nts.1814 to 1901) contains the start site for synth...

متن کاملRisk for hepatocellular carcinoma with respect to hepatitis B virus genotypes B/C, specific mutations of enhancer II/core promoter/precore regions and HBV DNA levels.
BACKGROUND/AIM To examine the risks for hepatocellular carcinoma (HCC) with respect to hepatitis B virus (HBV) genotypes, specific viral mutations (MT), serum HBV DNA levels, and cirrhosis. METHODS HBV genotypes, 1653/1753/core promoter (CP)/precore MT and HBV DNA levels were determined in 248 HBV patients with HCC and 248 HBV controls. RESULTS Genotype C, CP-MT, T1653, HBV DNA levels >or=4...
